چکیده

sequence encoding the chloroplast RecA protein was identified from the Kazusa EST database for the unicel)ular green alga, C7lalamydomonas reinhardtii (http:orwww.kazusa.or.jp/en/plant!chlamy!ESTI). Analysis of the cDNA sequence identified an open reading frame (ORF) of 414 amino acids encoding a eubacteria-type RecA protein. Thus the corresponding gene was named RECI. The predicted protein contains an N-terminal extension that does not show any similarity with other RecA proteins. Transient expression of a RECI-sGFP (green fiuorescent protein) fusion construct in tobacco cells has indicated that this N-terminal sequence functions as a transit peptide for import into

Expression and complementation analyses of a chloroplast-localized homolog of bacterial RecA in the moss Physcomitrella patens.

RecA protein is widespread in bacteria, and it plays a crucial role in homologous recombination. We have identified two bacterial-type recA gene homologs (PprecA1, PprecA2) in the cDNA library of the moss Physcomitrella patens. N-terminal fusion of the putative organellar targeting sequence of PpRecA2 to the green fluorescent protein (GFP) caused a targeting of PpRecA2 to the chloroplasts. Muta...

Inhibition of chloroplast DNA recombination and repair by dominant negative mutants of Escherichia coli RecA.

The occurrence of homologous DNA recombination in chloroplasts is well documented, but little is known about the molecular mechanisms involved or their biological significance. The endosymbiotic origin of plastids and the recent finding of an Arabidopsis nuclear gene, encoding a chloroplast-localized protein homologous to Escherichia coli RecA, suggest that the plastid recombination system is r...

Genomic nucleotide sequence of a gene from Arabidopsis thaliana encoding a protein homolog of Escherichia coli RecA.

In Escherichia coli and many other prokaryotes, the RecA protein is essential for homologous recombination and for a variety of S.O.S. responses to DNA damage (Little and Mount, 1982; Kowalczykowski, 1991). Previously, Cerutti et al. (1992) found a chloroplast-localized strand recombination activity resembling that of bacterial RecA and a protein band cross-reacting with antibody to E. coli Rec...

Organisms must have efficient mechanisms of DNA repair and recombination to prevent alterations in their genetic information due to DNA damage. There is evidence for DNA repair and recombination in plastids of higher plants, although very little is known at the biochemical level. Many chloroplast proteins are of eubacterial ancestry, suggesting that the same could be true for the components of ...

Mechanisms for maintenance, replication, and repair of the chloroplast genome in plants.

Photosynthesis is a complex process that occurs in chloroplasts of higher plants, and requires a large number of proteins to assemble the photosynthetic machinery. Many chloroplast-localized proteins are nuclear-encoded and must be imported into the chloroplasts from the cytoplasm. A considerable number of genes for photosynthesis and other chloroplast functions, including transcription and tra...
